Key words myelodysplastic syndromes ;aenic sulfide; BMMNC; apoptosis; bcl-2; bax 骨髓增生异常综合征(myelodysplastic syndromes，MDS)是一种造血干细胞异常克隆性与异质性疾病，其主要特征为病态造血和无效造血，部分患者易向白血病转化。MDS发病率有逐年上升趋势［1］，但目前对其 治疗 仍无很好的方法。经研究发现，砷剂具有致癌、致畸、致突变的协同作用；作为微量元素，砷尚能促进造血、细胞生长和繁殖［2］。硫化砷（As2S2）是中药雄黄的主要成分，近年来，用雄黄治疗MDS取得了一定的疗效［3］，本研究以观察硫化砷（As2S2）在体外对MDS骨髓单个核细胞的作用，查明其可能的治疗机理，开发其应用领域。 材料和方法 病例选择 MDS患者25例，按照FAB分型［4］，RA 15例，RAS 3例，RAEB 5例，RAEB-t 2例。男性16例，女性9例，中位数年龄45（19-80）岁，为初诊及复诊的患者，每例均经骨髓细胞形态学和骨髓病理等相关的实验室检查确诊。以上病例来自南通大学附属 医院 血液科。 标本采集和细胞分离 自髂后上棘抽取5 ml骨髓液，%肝素抗凝，采集后2小时内用淋巴细胞分离液密度梯度离心分离骨髓单个核细胞，RPMI 1640培养液洗3遍，制成细胞悬液，计数备用。 试剂 硫化砷（As2S2）购自美国Sigma公司，纯度%，以 mol/L NaOH 溶解，HCl中和滴定至pH
