CD73isaglycosyl-phosphatidylinositol-(GPI-)linkedmembraneproteinthatcatalyzestheextracellulardephosphorylationof adenosinemonophosphate(AMP)toadenosine.Adenosineisanegativeregulatorofin?ammationandpreventsexcessivecellular damage.WeinvestigatedtheroleofextracellularadenosineintheintestinalmucosaduringthedevelopmentofDextran-Sulfate- Sodium-(DSS-)salt-inducedcolitisinmicethatlackCD73(CD73?/?)andareunabletosynthesizeextracellularadenosine.We havefoundthat,comparedtowild-type(WT)mice,CD73?/?micearehighlysusceptibletoDSS-inducedcolitis.CD73?/? mice exhibitpronouncedweightloss,slowerweightrecovery,anincreaseingutpermeability,adecreaseinexpressionoftightjunctional adhesionmolecules,aswellasunresolvedin?ammationfollowingtheremovalofDSS.Moreover,colonicepitheliainCD73?/? miceexhibitedincreasedTLR9expression,highlevelsofIL-1βandTNF-α,andconstitutiveactivationofNF-κB.Weconcludethat CD73expressioninthecoloniscriticalforregulatingthemagnitudeandtheresolutionofcolonicimmuneresponses. 1.Introduction and their associated antigens, must be kept isolated from intraepitheliallymphocytestoavoidabnormala
