CD8TcellsareessentialforcontrolofToxoplasmagondiiinfection.Onceactivatedtheyundergodi?erentiationintoshort-lived e?ectorandmemoryprecursore?ectorcells.Ase?ectorcells,CD8Tcellsexertimmunepressureontheparasiteviaproduction ofin?ammatorycytokinesandthroughtheircytolyticactivity.Onceimmunecontrolhasbeenestablished,theparasiteencysts anddevelopsintochronicinfectionregulatedbythememoryCD8T-cellpopulation.Severalsignalsareneededforthisprocess tobeinitiatedandfordevelopmentoffullydi?erentiatedmemoryCD8Tcells.WithnewlydevelopedtoolsincludingCD8T- celltetramersandTCRtransgenicmice,dissectingthebiologybehindT.gondii-speci?cCD8T-cellresponsescannowbemore e?ectivelyaddressed.Inthispaper,wediscusswhatisknownaboutthesignalsrequiredfore?ectiveT.gondii-speci?cCD8T-cell development,theirdi?erentiation,ande?ectorfunction. 1.Introduction were important for control of infection with CD8 T cells playingthedominantrole[6].Furtherstudiesbythisgroup usingasimilarstrategywithanti-IFNγandadoptivetransfer of CD8T cells identi?ed that IFNγ was a major mediator of disease [7]. A short time later Khan and colleagues developedthe?rstantigen-speci?cCD8Tcellclonescapable of responding and killing T. gondii tachyzoites in vitro via theircytotoxicactivity[8].Thesestudiessuggestedthattwo e?ectormechanismsmaybeatplayincontrollingT.gondii infection
