Heatshockproteins(HSPs)havebeenknownfordecadesfortheirabilitytoprotectcellsunderstressfulconditions.Inthe1980sa newrolewasascribedforseveralHSPsgiventheirabilitytoelicitspeci?cimmuneresponsesinthesettingofcancerandinfectious disease.Theseimmuneresponseshaveprimarilybeenharnessedfortheimmunotherapyofcancerintheclinicalsetting.However, becauseoftheabilityofHSPstoprimediverseimmuneresponses,theyhavealsobeenusedformodulationofimmuneresponses duringautoimmunity.TheapparentdichotomyofimmuneresponseselicitedbyHSPsisdiscussedhereonamolecularandcellular level.ThepotentialclinicalapplicationofHSP-mediatedimmuneresponsesfortherapyofautoimmunediseasesisreviewed. 1.Introduction:HSPsinImmunity receptor has now been shown to be CD91. These events arediscussedinthenexttwosubheadings.Theisolationof HSPs(andtheassociatedpeptides)fromtumorcellsorcells infected with pathogens therefore provides a single entity thatprimesimmuneresponsesspeci?cforthechaperoned peptidesandthusforthecellthatharboredtheseantigens. Thisapplicationhasbeentestedinavastnumberofrodent modelsofcancerandinfectiousdiseaseandisbeingtestedin theclinicalsetting.Wediscussthisinthethirdsubheading. AsearchforoptimalimmunizingdosesofHSPsledtothe fortuitous dampening of the immune response at higher doses of HSPs. This phe
