Recently,ithasalsobeenimplicatedinTh2-mediatedallergicdiseases,suchasasthma.TheexpressionofOPNin allergicrhinitis(AR)iscurrentlyunknown,asistheeffectofintranasalglucocorticosteroids(GCs)onthatexpression. Methods:SubjectswithARwererandomisedtoreceivetreatmentwithfluticasonepropionate(FP)(n=12)ora placebo(n=16)overthegrasspollenseasonandnasalbiopsiesweretakenpriorto,andduringtheseason.OPN expressioninthenasalmucosawasexaminedwithimmunohistochemistry.Healthynon-ARcontrols(n=5)were usedasacomparator. Results:OPNexpressionwasdetectedinepithelialcells,subepithelialinfiltrating/inflammatorycellsandcellslining thevesselsandglandsofallsubjects.Comparisonofthepre-andpeak-pollenseasonbiopsysectionsinplacebo treatedpatientsrevealednoincreaseinOPNexpressionduringthegrasspollenseason(5.7%vs6.4%).Treatment withalocalglucocorticosteroiddidnotaltertheexpressionofOPNduringpollenexposure(6.2%vs6.7%). Conclusion:OPNhasbeenincreasinglyassociatedwiththepathogenesisofvariousTh2-mediateddiseases. However,ourfindingthattheOPNexpressioninthenasalmucosaofARpatientsisnotsignificantlyaffectedby allergenexposureandiscomparabletothatofthehealthycontrols,suggeststhatintracellularOPNisnotdirectly involvedinthepathogenesisofallergicrhinitis.
