RSFpossessesnotonlychromatinremodelingactivitybutalsochromatinassemblyactivityinvitro.Whilenoothersinglefactor canexecutethesameactivitiesasRSF,thebiologicalsignificanceofRSFremainedunknown.Toinvestigatetheinvivofunction ofRSF,wegeneratedamutantalleleofDrosophilaRsf-1(dRsf-1).ThedRsf-1mutantbehavedasadominantsuppressorof positioneffectvariegation.IndRsf-1mutant,thelevelsofhistoneH3K9dimethylationandhistoneH2AvariantH2Avwere significantly reduced in an euchromatic region juxtaposed with heterochromatin. Furthermore, using both genetic and biochemicalapproaches,wedemonstratethatdRsf-1interactswithH2AvandtheH2Av-exchangingmachineryTip60complex. TheseresultssuggestthatRSFcontributestohistoneH2Avreplacementinthepathwayofsilentchromatinformation.
