MicroRNAs,aclassofsmallnon-codingRNAs,havebeenimplicatedinthepathogenesisandprognosisofcolorectalcancer, althoughfewstudieshaveexaminedtherelationshipbetweengermlinemutationinthemicroRNAswithriskandprognosis. WethereforeinvestigatedtheassociationbetweenaSNPinhsa-mir-608,whichlieswithinthe10q24locus,andcolorectal cancer. MethodsandResults:Acohortconsistingof245casesand446controlswasgenotypedforrs4919510.Thefrequencyof theGGgenotypewassignificantlyhigherinAfricanAmericans(15%)comparedtoCaucasians(3%)controls.Therewasno significantassociationbetweenrs4919510andcolorectalcancerrisk(AfricanAmerican:ORGG vs. CC0.89[95%CI,0.41–1.80]) (Caucasian:ORGG vs. CC1.76,([95%CI,0.48–6.39]).However,wedidobserveanassociationwithsurvival.TheGGgenotype wasassociatedwithanincreasedriskofdeathinCaucasians(HRGG vs. CC3.54([95%CI,1.38–9.12])andwithareducedriskof deathinAfricanAmericans(HRGG vs. CC 0.36([95%CI0.12–1.07). Conclusions:Theseresultssuggestthatrs4910510maybeassociatedwithcolorectalcancersurvivalinamannerthatis dependentonrace.
