Methods:Prospectiveanalysiswasperformedofbankedtumorandserumspecimensfrom99patientswith thyroidmasses.Enzyme-linkedimmunosorbentassay(ELISA)wasemployedtomeasurelevelsoffiveserum proteinspreviouslydemonstratedtobeup-regulatedinpapillarythyroidcancer(PTC):angiopoietin-1(Ang-1), cytokeratin19(CK-19),tissueinhibitorofmetalloproteinase-1(TIMP-1),chitinase3like-1(YKL-40),andgalectin-3 (GAL-3).SerumlevelswerecomparedbetweenpatientswithPTCandthosewithbenigntumors. Results:Atotalof99patientswereenrolledinthestudy(27men,72women),withamedianageof54years. Forty-threepatientshadPTCand58caseswerebenigntumors.Therewerenostatisticallysignificantdifferences whencomparingallfivedifferentbiomarkersbetweenPTCandotherbenignthyroidtumors.Thep-valueswere 0.94,0.48,0.72,0.48,and0.90forYKL-40,Gal-3,CK19,TIMP-1,andAng-1,respectively. Conclusion:Serumlevelsoffourofthefiveproteinswereelevatedinpatientswiththyroidmassesrelativeto normalvalues.However,thedifferencebetweenbenignandPTCwasnotsignificant.Twoofthemarkers(Gal-3 TIMP-1)displayedagreaterpotentialdifference,whichmaywarrantfurtherinvestigation.Thisstudysuggeststhat otherserummarkersshouldbesought.Thisisthefirststudytoinvestigatepotentialserumbiomarkersbasedon over-expressedproteinsinthyroidcancerversusbenignpathology.
