ShiftintheIntrinsicExcitabilityofMedialPrefrontal CortexNeuronsfollowingTraininginImpulseControl andCued-RespondingTasks ScottJ.Hayton1,MaryC.Olmstead1,2. ′ ,EricC.Dumont 2,3 . * 1Department of Psychology, Queen’s University, Kingston, Ontario, Canada, 2Centre for Neuroscience Studies, Queen’s University, Kingston, Ontario, Canada, 3DepartmentofAnesthesiologyandPerioperativeMedicine,Queen’sUniversity,Kingston,Ontario,Canada Abstract Impulsecontrolisanexecutiveprocessthatallowsanimalstoinhibittheiractionsuntilanappropriatetime.Previously,we reportedthatlearningasimpleresponseinhibitiontaskincreasesAMPAcurrentsatexcitatorysynapsesintheprelimbic regionofthemedialprefrontalcortex(mPFC).Here,weexaminedwhethermodificationstointrinsicexcitabilityoccurred alongside the synaptic changes. To that end, we trained rats to obtain a food reward in a response inhibition task by withholdrespondingonaleveruntiltheyweresignaledtorespond.Wethenmeasuredexcitability,usingwhole-cellpatch clamprecordingsinbrainslices,byquantifyingactionpotentialsgeneratedbytheinjectionofdepolarizingcurrentsteps. TraininginthistaskdepressedtheexcitabilityoflayerVpyramidalneuronsoftheprelimbic,butnotinfralimbic,regionof the mPFC relative to behavioral controls. This decrease in maximum spiking frequency was significantly correlated with performanceonthefinalsessionofthetask.Thischangeinintrinsicexcitabilitymayrepresentahomeostaticmechanism counterbalancingincreasedexcitatorysynapticinputsontothoseneuronsintrainedrats.Interestingly,subjectstrainedwith acuethatpredictedimminentrewardavailabilityhadincreasedexcitabilityininfralimbic,butnottheprelimbic,pyramidal neurons. This dissociation suggests that both prelimbic and infralimbic neurons are involved in directing action, but specializedfordifferenttypesofinformation,inhibitoryoranticipatory,respectively.
