Acta Physiologica Sinica，December 25，2007，59(6)：840-844 http：／／www．actaps．corn．cn Electrophysiologic effects of 17 diol on pacemaker cells in sinoatrial 1 ● ■ ’ ● · noaes OI-raDDits ZHOU Li—Bin，SONG Li—Lin，GUAN Yue，GUO Shu—Mei，YUAN Fang，ZHANG Yi Department ofPhysiology,Hebei Medical University,Shijiazhuang 05001 7,China Abstract：To investigate the electrophysiological effects of l 7p—estradiol on pacemaker cells in sinoatrial(SA)nodes of rabbits and the underlying mechanism，intracellular microelectrode technique was used to record action potential(AP)in SA node cells of rabbits．The results showed that：(1)17p—estradiol(1，10，100 lamol／L)not only significantly decreased the amplitude of action potential(APA)and the maximal rate of depolarization( )，but also decreased the velocity of diastolic(phase 4)depolarization(VDD)and rate of pacemaker firing(RPF)in a concentration—dependent manner．The AP duration at 50％repolarization(APD50)and at 90％repolariza— tion(APD90)were prolonged．But the maximal diastolic potential(MDP)was not affected．(2)Pretreatment with tamoxifen(10 gmol／ L)，an inhibitor of estrogen receptor，did not block the electrophysiological effects of 17B—estradiol(10 gmol／L)on SA node cells．(3) Pretreatment with 一nitro—L—arginine methyl ester(L—NAME，100 gmol／L)，a nitric oxide(NO)synthase inhibitor，completely abolished the elec仃ophysiological effects of 17p—estradiol(10 lamol／L)on SA node cells．The results suggest that 17B—estradiol inhibits the electrophysiological activity of pacemaker cells in SA nodes of rabbits in a concentration—·dependent manner possibly through a non—． genomic mechanism related with NO． Key words：17B—estradiol；sinoatrial node；action potential；tamoxifen；L—NAME 雌二醇对家兔窦房结自律细胞的电生理学效应 周利彬，宋立林，关明，郭书梅，袁芳，张翼 河北医科大学生理学教研室，石家庄
