strokeinaratmodel,themodelisbuilttosuccessfullyevaluatethemorphology,pathology,andtheexpressionofHIF-1αintheratbrainwithitssignificance.Methods60healthyadultmalewistarrats,inthecontrolgroup(n=30)andexperimentalgroup(n=30),preparedonthebasisofhealthyratmodelofischemicstrokeandatherosclerosisinratswithischemicstrokeonthebasisofModel.Eachgroupisdividedintoshamoperation,ischemia0.5,6,12,24hsubgroup.ThesuccessofthemodelingisdeterminedbythemorphologyobservedunderlightmicroscopeHEstainingandpathophysiology.ThechangesinexpressionofHIF-1αmRNAaredetectedbyimmunohistochemicalexpressionofHIF-1αproteinandRT-PCR.Measureddataareanalyzedbyimageanalysissystem,whichmeasurestheaverageproteinandmRNAexpressioninopticaldensity,theaveragegrayvalue.Thedataarealsoanalyzedbyspss17.0forstatisticalanalysis.ResultsThesuccessfullyofthemodelsymbolofsuccessofmodelofatherosclerosis:HEstaining,lightmicroscopyofaorticendothelialdiscontinuity,plaqueformation;calcification;inthesmoothmusclehyperplasia,elasticfiberbreakage;outermembranethinning，symbolofsuccessmodelofischemicstroke:aftertheobservationofanimalbehaviorandbodyawakening,globalcerebralischemiamydriasis,conjunctivaldarkred,archedverticalhair,surgicalsuccess.Immunestaining:0.5hcontrolgroupandexperimentalgroupinthesubgroupofischemicbraintissueHIF-1αexpressionhasbeenobvious,6h,reachedthepeaksub-group,12,24hinthecontinuedhighexpressionsubgroup.Statisticallysignificant:experimentalgroupandcontrolgroupwithshamoperationsubgroup0.5hsub-groupwasstatisticallydifferent(P0.05).6hsub-groupthaninsham,ischemia0.5hsignificantlydifferentsubgroups(P0.05).Ischemia12,24hsubgroups,respectively,comparedwith6hsubgroup,thereisnosignificantdifferencestatistically(P0.05).Nosignificantdifferencebetweentheexperimentalgroupandcontrolgroup.RT-PCRDetection:0.5hcontrolgroupandexperimentalgroupinthesubgroupofischemicbraintissuehasbeensignificantHIF-1αmRNAexpression,6h,reachedthepeaksub-group,12,24hinthecontinuedhighexpressionsubgroup，Statisticallysignificant:experimentalgroup
