色素痣石蜡标本中基因和蛋白水平的检测，得出的结论与脂溢性角化病相似。(3)两种HPVLI抗体的阳性表达部位的不同，提示抗体的差异，同时显示皮肤标本中角化层与未角化层均存在HPVLI蛋白的表达，至于两种抗体为何定位不同值得进一步探讨；两种抗HPVL1蛋白抗体检测两种病变组织HPVL1的表达情况，实验结果经过比较可证实本课题组研制的兔抗检测能力可与购买的鼠抗相当，。关键词：人乳头瘤病毒脂溢性角化病色素痣检测AbstractObjective：Theaimofthisresearchprojectistocollecttheclinicalspecimensofseborrheickeratosisandpigmentednevusbasedonpreviousstudies。ThenextracttotalDNA，todetectsomespecimensoftypeHPVDNAexistsornotinthesespecimensusingPCRofHPVspecialanduniversalprimerpairs。TodetecttheexpressofHPVcapsidproteinL1inparaffinofSKandpigmentednevus，usingimmunofluorescencetechniqueoftwoantibodiesfortheHPVL1protein。FromtwolevelsofgeneandproteintoinvestigatethepresenceofHPVinthetwoskinlesions，andtherelevanceofHPVandtheselesions。Methods：(1)Collectedtheparaffinspecimensofseborrheickeratosisandpigmentednevus，todeterminetheclinicaldiagnosisandpathologicaldiagnosis。Thesamesampleweredividedintotwoparts，oneismadeparaffinsectionof4umthick。Theotherismadeparaffinsectionof10umthickserialslices，take5tabletsputin1.5mlEPtube；(2)ToextracttotalDNAofspecimens，thendetectdethepresenceofHPVgenegenesornotbyusingPCR，tosurethepositiverateofHPVDNA；(3)TodetecttheexpressofHPVcapsidproteinL1inthesameparaffin，usingimmunofluorescencetechniqueofmouseanti-papillomavirusmonoclonalantibodyandrabbitanti-papillomavirusmulticlonalantibody，tosurethepositiverateofHPVL1protein。(4)Usingthestatisticalsoftware，comprehensiveanalysisofexperimentalresults，toconfirmthepresenceofHPVinseborrheickeratosisandpigmentednevus，anddiscusstheroleofHPVintheabovediseasepathogenesis。Results：(1)Thereare46paraffinspecimensofSK，pigmentednevusare52；(2)Afterβ－globingeneamplification，38casesofSKwerepositive，whilepigmentednevusare44，pickoutthepositivesamplesforHPVL1universalprimerMY09/11、GP5+/6+，HPV16E6，HPV18E6amplificationfortwice，PCRrevealedthattherewasoneof43SKsamplescontainedHPV16DNA，thedetectionratewas2.63%，therestwerenegative；(3)SKspecimensweredetectedbyimmunofluorescence，whenusingrabbitanti-papillomavirusmulticlonalantibody，positivepartmainlyinnon
