Abstract*Vesiculartraffickingisofcriticalimportanceforavarietyoflifefunctions,andtightlyrelatedtosomediseases.Itconsistsofexocystosisandendocytosis,whichareregulatedbymanymolecules.Studiesindifferentmodelorganismshaverevealedlotsofcomponentsinvolvedinvesiculartrafficking,butthemechanismofsomecomponentsisstillunclear.Inthispaper,wededicatetoexploringthemolecularmechnismofSEC-10andSynaptotagminVII（SytVII）inexocytosisandendocytosisrespectively.Especially,thestudyofSEC-10inCaenorhabditiselegans(C.elegans)combinedthecellulartrafficwithbehaviorcharacteristic,whichindicatesthenewroleofexocystinendocyticpathway.Inthisstudy,weinvestigatedtherolesofSytVIIonexocytosisoflargedensecorevesicles(LDCVs)inPC12cells.SytVII,whichhasahigherCa2+affinityandslowerdisassemblykineticswithlipidthanSytIandSytIX,wasregardedasbeinguninvolvedinsynapticvesicle(SV)exocytosisbutInsteadpossiblyasacalciumsensorfortheslowerkineticphaseofdensecorevesicles(DCVs)release.ButatpresentthelocalizationandfunctionofSytVIIremainunclear.UsingTIRFM,ourresultsshowedthatSytVIIispredominantlylocatedonDCVsinPC12cells.Byusinghightemporalresolutioncapacitanceandamperometrymeasurements,itwasdemonstratedthattheknockdownofendogenousSytVIIattenuatedthefusionofDCVwiththeplasmamembrane,reducedtheamplitudeoftheexocytoticburstoftheCa2+-triggeredDCVreleasewithoutaffectingtheslopeofthesustainedcomponent,andblockedthefusionporeexpansion.ThissuggeststhatSytVIIistheCa2+sensorofDCVfusionmachineryandisanessentialfactorfortheestablishmentandmaintenanceofthepoolsizeofreleasableDCVsinPC12cells.TheresearchesaretakenonC.eleganstorevealtherolesofSEC-10inworm*ThisworkwassupportedbygrantsfromtheMajorResearchplanfromtheMinistryofScienceandTechnologyofChina(No.2007CB914200),theNationalScienceFoundationofChina(No.behaviorsandkindsoflifeaffairs.Especially,westudythemolecularmechanismofSEC-10inendocytoticpathwayofkindsofcells.Sec10isonecomponentofexocyst(Sec3,Sec5,Sec6,Sec8,Sec10,Sec15,Exo70,Exo84),whichisstronglyimplicatedincellulartra
