AbstractBotrytis cinerea is one of the major postharvest pathogens in plants, it can cause great heavy economic losses. Nitric oxide (NO) is a kind of unique and signaling molecule, playing an important role in cells. It has both toxicity and protective functions, dependenting on the changes of concentration, which means higher concentrations inhibit cell growth and accelerate cell apoptosis. Nowadays some studies have showed that exogenous NO can inhibit the growth of pathogens. In this study, we investigated the different concentrations of NO if it could inhibit spore germination of the postharvest pathogen B. cinerea systematically. We explore the possible mechanisms by cell biology, biochemistry and molecular biology. The main results are as followings:The antifungal experiments showed that nitric oxide donor SNP can significantly inhibit the growth of B. cinerea at 4 mmol/L, and the effect of inhibitory is dependent on dose and time.Through the propidium iodide staining, we analysis that 3 mmol/L NO treatment can cause extracellular soluble carbohydrate and protein increased. The results confirmed that NO can lead to damage of the membrane, resulting in inhibiting the growth and development of B. cinerea.Through dichlorofluorescein fluorescence probe (DCHF-DA) detection, we found that SNP at 4 mmol/L could increase the level of reactive oxygen species (ROS). In addition, the spore intracellular antioxidant system was destroyed, antioxidant enzyme activity and antioxidants content was decreased, these results indicated that NO induces the generation of ROS.NO could effectively delay pericarp turning to red of tomato fruit, delay respiratory, and reduce the intensity of respiration. In addition, NO could maintain the high level of antioxidant enzyme activity and keep the good quality of fruits, enhance the disease resistance of fruit.Keywords: NO Botrytis cinerea ROS oxidative damage目录摘要 .........................................................................
