⑧————』型竺堡些塑L——一摘要本论文将生物合理设计方法与平行液相组合合成有机结合起来，进行新农药 的创制，以期发现新的先导化合物，同时对重氮化合物的氢取代反应、以及5一 氨基．3．甲硫基．IH．毗唑的甲基化反应也进行了研究。吡唑连吡唑类化合物是 类高效的除草剂，其生物活性的体现与其耿代基有 着密切联系。为了更好的了解吡唑连吡唑化合物的构效关系以及发现新的先导化 合物，我们将传统有机合成与平行液相组合合成有机结合，合成了含有23个子 库的吡唑连吡唑化合物库。每个化合物子库均经过核磁共振、元素分析和液谱或 GCD确证。设计并合成了吡唑并[1，5-a]嘧啶类化合物，其结构经核磁共振、元素分析确证。制备了其中两个化合物的单晶，并经x．衍射分析。 设计并合成了6个新型毗唑腙类化合物，其结构经核磁共振、红外光谱、元素分析确证。在吡唑重氮化合物的氢取代反应中，我们偶然发现铜有着非常重要的作用， 进而详细研究了在铜存在时，温度、溶剂、时阿等因素对反应的影响。实验表明， 在有铜存在的条件下反应速率加快、反应产率提高。当5．氨基．3．甲硫基．1H．吡唑进行甲基化反应时，吡唑环上的两个N原子都 可能有甲基取代，从而生成～对异构体。在实验过程中，当4-位取代基分别为氰 基和乙氧羰基时，两个反应中生成的异构体的比例相差甚远，为了找到影响区域 选择性的因素，更好地了解吡唑的甲基化反应，我们对5一氨基一3一甲硫基-1H一吡 唑的甲基化反应进行了细致的研究，部分产物经x一射线单晶衍射确证结构，并 利用从头计算对电子效应进行了研究。对合成的部分化合物进行了初步的生物活性测定，研究发现大部分化合物都表现出一定的除草活性。ⅡI⑩南开大学博士学位论文关键词：吡唑连吡唑类化合物，先导化合物，平行液相组合合成，化合物库，甲基化 反应，靶酶抑制剂，重氮化合物，x-射线衍射，从头计算，生物活性1V⑧————·堡型兰唑型墨———一ABSTRACTThe methods of bionational drug design and parallel-liquid·-phase combinatorial synthesis were combined together as an approach to the preparation of combinatorial libraries containing pyrazole derivatives and to the discovery of new lead compounds． Moreover the reaction of diazo pyrazoles and the methylation of 5-amino一3一methylthio-1H-pyrazole derivatives were also studied．Prompted by the observation that many pyrazolylpyrazole derivatives are used as herbicides．A small combinatorial library containing pyrazolylpyrazole derivatives was synthesized by traditional organic synthesis and parallel·-liquid-phase combinatorial synthesis．New lead compounds were expected to be discoveried through this library．The structures of compounds were confirmed by 1HNMR、LC and elementary analysis．pyrazolo[1，5-a]pyrimidine compounds were synthesized，and their structures were confirmed by 1HNMR and elementary analysis。Single crystals oftwo ofthose compounds Were Obtained and their structures were confirmed further by X·ray analyses．Six pyrazole hydrazonesweredesignedand synthesized，and theirstructureswere confirmed by 1HNMR、IR and elementary analysis。It was found accidentally that copper
