山西医 山西医科大学硕士学位论文 I I 脑出血后 ephrin—B3 的表达与白质损伤的相关性 摘要 目的；运用elisa技术对成年大鼠脑出血后ephrin-b3的表达进行定量测定， 进而探讨成年大鼠脑出血（ICH，Intracerebral hemorrhage）后ephrin-b3的表达 与脑白质损伤的关系及其作用机制，为脑出血后神经纤维的再生提供新的思路与 技术方法。 方法；采用250+15g的wistar雄性清洁级大鼠56只，随机分为正常对照组（n=8 只），假手术组（n=8只），出血组（n=40只）。大鼠麻醉后，在立体定位仪辅助仪 器下，取大鼠尾动脉非肝素不凝血100 ul，分3次注入内囊区以建立脑出血模型， 分别于造模成功后6，24,48,72h,7d处死，断头取脑。应用苏木精-伊红染色观察 各组血肿周围脑组织的病理变化；应用elisa法检测对照组及出血组血肿周围 ephrin-b3的表达情况。 结果：光镜（HE 染色）病理观察：出血6h 组于血肿周围可见变性坏死区域， 24h、48h、72h组血肿周围的变性坏死区逐渐扩大，72h损伤达高峰， 7d与72h组 无明显变化。elisa法检测血肿周围白质ephrin-b3的表达：出血后6h血肿周围 ephrin-b3的表达开始升高，24h,48h表达逐步增高，72h表达达到高峰，它们之间 两两比较（p0.05），具有统计学意义；与相应的时间点的假手术组和正常组两两 比 较 （ p0.05 ）， 有 统 计 学 意 义 ； 出 血 7d 组 ephrin-b3 表 达 恢 复 明 显 ， 与 6h,24h,48h,72h组两两比较（P0.05），有统计学意义；与假手术组和正常对照组 两两比较虽有差异，但（p0.05）无统计学意义；假手术各组和正常对照组比较 （p0.05）,无统计学意义。 结论：成年大鼠脑出血后 ephrin-b3 的表达和脑白质损伤程度相一致，提示 ephrin-b3 可能参与了脑出血损伤的发生，在脑出血发生、发展和转归过程中起 一定的作用。 关键词：脑出血；ephrin-B3；再生；脑白质损伤； I II The revelance of ephrin-b3 expression and white matter damage after intracerebral hemorrhage Abstract Objectives：To explore the revelance of ephrin-b3 expression and white matter damage and the mechanism of it in intracerebral hemorrhage adult rat. Methods: 250+15g male Wistar rats were used.developing the not coagulation which was taken from the rat tail and injecting it to the brain internal capsule to establish the model of intracerebral hemorrhage.Kill the rat and take the brain at 6,24,48,72h,7d point after the success of model,meanwhile establish the appropriate control group.Application of hematoxylin-eosin staining to observe the brain tissue around the homatoma pathological changes.Application of elisa assay to detect the expression of ephrin-b3 around the hematoma. Results：Compared with the control group ,6h、24h、48h、72hthe necrotic area around the hematoma gradually expanded and infiltration of lymphocytes and neutrophils.7d and 72h group with no significant change.
