上转换纳米探针在循环肿瘤细胞富集与检测中的应用中文摘要中文摘要外周血中循环肿瘤细胞的出现是肿瘤转移和复发的重要指征。然而在肿瘤患者的外周血中循环肿瘤细胞的密度极低，大约平均106个细胞中仅有一个肿瘤细胞。因此，针对循环肿瘤细胞的高效捕获和检测方法在肿瘤早期诊断和预后评估中起着极为重要的作用。然而高灵敏度和高特异性的且重复性好的检测方法仍有待于进一步地研究与开发。本课题首次提出将上转换发光材料结合核酸适配体作为纳米探针来检测循环肿瘤细胞，并在磁场作用下，利用磁性纳米材料富集检测到的循环肿瘤细胞。我们首先将修饰的UCNPs结合生物素，同时接上特异性识别靶细胞蛋白酪氨酸激酶PTK-7的核酸适配体，得到可特异性识别靶细胞(CCRF-CEM细胞)的Biotin-UCNPs-Aptamer复合物。然后用修饰了亲和素的磁性纳米材料来分离靶细胞，便于靶细胞的进一步分析。由于上转换材料UCNPs无自发性荧光的特性以及磁性纳米材料对靶细胞的有效分离，避免了检测过程中背景荧光信号的干扰，也大大降低了检测分离中的非特异性吸附，使这一新型检测方法在标准样本的检测中有潜力达到高灵敏检测和高捕获率的功效。研究结果显示，在模拟的临床样本的检测中，即使只有10个靶细胞，使用我们的检测模型也能达到较高的捕获效率，而且富集的细胞有着较高的活性，以利于对靶细胞进一步的分析。因此，我们开发的新方法对于CTCs的检测和分离，是一个有潜力的，具有高特异性和灵敏度的且简便可行的方法，对未来肿瘤临床诊断和肿瘤患者术后预防治疗具有重大的意义。关键词：上转换材料，循环肿瘤细胞，磁性纳米材料，核酸适配体作者：方帅指导老师：彭睿AbstractUpconversionnanoprobesappliedineffectivecaptureandsensitivedetectionofcirculatingtumorcellsUpconversionnanoprobesappliedineffectivecaptureandsensitivedetectionofcirculatingtumorcellsAbstractCirculatingtumorcells(CTCs)shedfromtheprimarytumorintobloodstreamisanimportantindicationofcancermetastasis.Theireffectivecaptureanddetectionarethereforeofgreatimportanceincancerdiagnosisandprognosis.However,duetotheextraordinarilyrarenumberofCTCsinthebloodofcancerpatients(onein106hematologiccells),thedevelopmentofnewCTCsdetectiontechnologythatiseasyoperationandhighlysensitivestillmeritsfurtherexplorations.Inthisstudy,aptamerconjugatedupconversionnanoparticles(UCNPs)areusedforthefirsttimeasnanoprobestorecognizetumorcells,whicharethenenrichedbyattachingwithmagneticnanoparticles(MNPs)andplacinginthepresenceofamagneticfield.Firstly,athiolatedaptamertargetingPTK-7over-expressedonavarietyofcancercells,togetherwithbiotinmolecules,wereco-conjugatedtoUCNPsusedfortumorcellrecognition.Then,weuseMNPsconjugatedwithavidintoseparateCTCs.Owingtotheauto-fluorescencefreenatureofupconversionluminescenceimaging,aswellasthemagneticseparationtofurtherreducebackgroundsignals,ourtechniqueallowsforhighlysensitivedetectionandcollectionofsmallnumbersoftumorcellsspikedintohealthybloodsamples.Astheresults,highlysensi
