成都中医药大学博士学位论文成都中医药大学博士学位论文 PAGE PAGE 4 Research on the combinatorial-Differentiation of Adult Stem Cell Inducted by Zuogui Wan and Biology Safety Huang Yong (Chengdu University of Traditional Chinese Medicine, Sichuan, Chengdu, 610075) 【Abstract】: Objective To investigate the controlled effect of ZuoGui Wan’s compatibility on differentiation of the bone marrow-derived adult stem cells (ASC).To search for the combinatorial-differentiation inducer of ASC by studying the separated recipes of ZuoGui Wan. To provide experimental data for the roll of TCM in regenerative medicine by studying the inducer’s biological characteristics and biology safety. Methods The bone marrow-derived ASC was used to analytical processing by the withdrawal of Zuo Gui Wan. The bone marrow-derived ASC was observed by cytochemistry, immunohistochemistry, chromosome analysis, two-layer agar gel precipitation test, chondrogenesis test of nude mouse and induce tumor test.The differentiated stem and progenitor cells’ biological characteristics and biology safety were observed by the multi-index method in vivo and in vitro, and the results could be objectively evaluated after comprehensive analysis. Results All drug groups could induce the differentiation of bone marrow-derived ASC into osteoblast-like cells, chondrocytes, neuron-like cells, glial-like cells and vascular endothelium-like cells. The ZuoGui WanⅠ(complete formula) mainly induced the differentiation of bone marrow-derived ASC into neuron-like cells, the ZuoGui WanⅡ(modificated formula that subtract GuiJiao andLuJiao) mainly induced the differentiation of bone marrow-derived ASC into osteoblast-like cells and chondrocytes. The neuron specific enolase(NSE) decreased after GouQi and ShanZhuYu had subtracted from the the ZuoGui WanⅡ, but the NSE increased after ShuDi had subtracted, The results showed GuiJiao, LuJiao, GouQi and ShanZhuYu could induce the differentiation of ASC into neuron-like cells, but ShuDi could inhibit(or adjust) t
