Methodology and Principal Findings: Forty-eight (36.861.0y), overweight (BMI 27.860.7kg/m2) participants were randomizedtofourgroupsfor6-months;Control:energyintakeat100%ofenergyrequirements;CR:25%calorierestriction; CR+EX:12.5%CRplus12.5%increaseinenergyexpenditurebystructuredexercise;LCD:lowcaloriediet(890kcal/d)until 15%weightreductionfollowedbyweightmaintenance.Bodycomposition(DXA)andtotaldailyenergyexpenditure(TDEE) over14-daysbydoublylabeledwater(DLW)andactivityrelatedenergyactivity(AREE)weremeasuredafter3(M3)and6 (M6)monthsofintervention.WeightchangesatM6were 21.061.1%(Control), 210.460.9%(CR),210.060.8%(CR+EX) and213.960.8%(LCD).AtM3,absoluteTDEEwassignificantlyreducedinCR(2454676kcal/d)andLCD(2633666kcal/d) butnotinCR+EXorcontrols.AtM6thereductioninTDEEremainedlowerthanbaselineinCRcal/d)andLCD cal/d) but reached significance only when CR and LCD were combined (2351683kcal/d). In response to caloric restriction (CR/LCD combined), TDEE adjusted for body composition, was significantly lower by 2431651 and 2240683kcal/datM3andM6,respectively,indicatingametabolicadaptation.Likewise,physicalactivity(TDEEadjusted forsleepingmetabolicrate)wassignificantlyreducedfrombaselineatbothtimepoints.ForcontrolandCR+EX,adjusted TDEE(bodycompositionorsleepingmetabolicrate)wasnotchangedateitherM3orM6. Conclusions:Forthefirsttimeweshowthatinfree-livingconditions,CRresultsinametabolicadaptationandabehavioral adaptationwithdecreasedphysicalactivitylevels.Thesedataalsosuggestpotentialmec
