Furtetal.BMCPlantBiology2012,12:70 /1471-2229/12/70 RESEARCH ARTICLE OpenAccess Quantitativeanalysisoforganelledistribution anddynamicsinPhyscomitrellapatens protonemalcells FabienneFurt1,KyleLemoi2,ErkanTüzel2andLuisVidali1* Abstract Background:Inthelastdecade,themossPhyscomitrellapatenshasemergedasapowerfulplantmodelsystem, amenableforgeneticmanipulationsnotpossibleinanyotherplant.Thismossisparticularlywellsuitedforplant polarizedcellgrowthstudies,asinitsprotonemalphase,expansionisrestrictedtothetipofitscells.Basedon pollentubeandroothairstudies,itiswellknownthattipgrowthrequiresactivesecretionandhighpolarizationof thecellularcomponents.However,suchinformationisstillmissinginPhyscomitrellapatens.Togaininsightintothe mechanismsunderlyingtheparticipationoforganelleorganizationintipgrowth,itisessentialtodeterminethe distributionandthedynamicsoftheorganellesinmosscells. Results:WeusedfluorescentproteinfusionstovisualizeandtrackGolgidictyosomes,mitochondria,andperoxisomes inliveprotonemalcells.Wealsovisualizedandtrackedchloroplastsbasedonchlorophyllauto-fluorescence.We showedthatinprotonemataallfourorganellesaredistributedinagradientfromthetipoftheapicalcelltothe baseofthesub-apicalcell.Forexample,thedensityofGolgidictyosomesis4.7and3.4timeshigheratthetipthan atthebaseincaulonemataandchloronematarespectively.WhileGolgistacksareconcentratedattheextremetip ofthecaulonemata,chloroplastsandperoxisomesaretotallyexcluded.Interestingly,caulonemata,whichgrow fasterthanchloronemata,alsocontainsignificantlymoreGolgidictyosomesandfewerchloroplaststhanchloronemata. Moreover,themotilityanalysisrevealedthatorganellesinprotonematamovewithlowpersistencyandaverage instantaneousspeedsrangingfrom29to75nm/s,whichareatleastthreeordersofmagnitudeslowerthanthose ofpollentubeorroothairorganelles. Conclusions:Toourknowledge,thisstudyreportsthefirstquantitativeanalysisoforganellesinPhyscomitrella patensandwillmakepossiblecomparisonsofthedistributionanddynamicsoforganellesfromdifferenttip growi
