Yamazakietal.BMCPhysiology2011,11:4 /1472-6793/11/4 RESEARCH ARTICLE OpenAccess SodiumselectivityofReissner’ smembrane epithelialcells MuneharuYamazaki1,2,KyungheeXKim1,DanielCMarcus1* Abstract Background:SodiumabsorptionbyReissner’smembraneisthoughttocontributetothehomeostasisofthe volumeofcochlearendolymph.Itwaspreviouslyshownthattheabsorptivetransepithelialcurrentwasblockedby amilorideandbenzamil.Themostcommonly-observedtargetofthesedrugsistheepithelialsodiumchannel (ENaC),whichiscomposedofthethreesubunitsa-,b-andg-ENaC.However,otherless-selectivecationchannels havealsobeenobservedtobesensitivetobenzamilandamiloride.Theaimofthisstudywastodetermine whetherReissner’smembraneepithelialcellscouldsupportparasensoryK+ absorptionviaamiloride-andbenzamil- sensitiveelectrogenicpathways. Results:Wedeterminedthemolecularandfunctionalexpressionofcandidatecationchannelswithgenearray (GEOGSE6196),RT-PCR,andwhole-cellpatchclamp.TranscriptexpressionanalysisofReissner’ smembrane detectednoamiloride-sensitiveacid-sensingionchannels(ASIC1a,ASIC2a,ASIC2b)noramiloride-sensitivecyclic- nucleotidegatedchannels(CNGA1,CNGA2,CNGA4,CNGB3).Bycontrast,a-,b-andg-ENaCwereallpreviously reportedaspresentinReissner’smembrane.Theselectivityofthebenzamil-sensitivecationcurrentswasobserved inwhole-cellpatchclamprecordingsunderCl--freeconditionswherecationsweretheonlypermeantspecies.The currentswerecarriedbyNa+ butnotK+,andthepermeabilityofLi+ wasgreaterthanthatofNa+ inReissner’s membrane.CompletereplacementofbathNa+ withtheinpermeablecationNMDG+ ledtothesameinward currentaswithbenzamilinaNa+ bath. Conclusions:Theseresultsareconsistentwiththeamiloride/benzamil-sensitiveabsorptivefluxofReissner’s membranemediatedbyahighlyNa+-selectivechannelthathasseveralkeycharacteristicsincommonwith abg-ENaC.Theamiloride-sensitivepathwaythereforeabsorbsonlyNa+ inthisepitheliumanddoesnotprovidea parasensoryK+ effluxroutefromscalamedia.
