RNA干扰Aurora A表达对人子宫内膜癌细胞的生长与细胞周期的影响 作者：滑芳 单位：石家庄市，河北医科大学第一医院妇产科 【摘要】 目的 探讨应用RNA干扰技术抑制Aurora A基因表达，并研究Aurora A基因表达下调对人子宫内膜癌细胞生长和细胞周期分布的影响。方法 用RNA干扰法抑制Aurora A的蛋白表达，RTPCR及Western blot方法检测Aurora A mRNA和蛋白表达,流式细胞仪检测细胞周期分布的变化,四氮唑盐法(MTT)检测转染前后细胞生长抑制率变化。结果 siRNA Aurora A可明显降低Aurora A mRNA和蛋白的表达,siRNA Aurora A导入Ishikawa细胞48 h后,细胞的生长被抑制，细胞周期阻滞于G2/M期和S期；细胞的凋亡率明显升高。结论 下调Aurora A表达可抑制人子宫内膜癌细胞系Ishikawa的增殖,同时导致细胞阻滞于G2/M期及S期并且促进细胞凋亡。 【关键词】 Aurora A激酶 子宫内膜癌 Ishikawa细胞 　　 The effect of siRNA Aurora A on cell growth and cell cycle in human endometrial carcinoma cell in vitro HUA Fang, ZHANG Hongzhen，FANG Guiying,et al. Department of Gynecology and Obstetrics, The First Hospital of Heibei Medical University, Shijiazhuang 050031 【Abstract】 Objective To address the possibility of Aurora A kinas as a therapeutic target for endometrial carcinoma, we employed siRNA technique to inhibit Aurora A expression and investegate its effect on tumor growth and cell cycle of Ishikawa cell.Methods Oligo siRNA of Aurora A was synthesized and transfected into Ishikawa cells. Aurora A mRNA expression and protein content were examined by reverse transcription polymerase chain reaction (RTPCR) and Western blot respectively. Cell cycle and apoptosis distribution were observed by flow cytometer. MTT assay was used to evaluate cell inhibition ratio before and after transfection. Results The aurora A mRNA expression and protein content significantly decreased by siRNA of Aurora A. The proliferation of the Ishikawa cells was inhibited by siRNA of Aurora A, along with the induction of accumulation of cells in S phase and the G2/M transition and increased cell apoptosis.Conclusion Inhibition of Aurora A expression can result in the suppression of cell growth and G2/M arrest of human endometrial carcinoma cell in vitro. 【Key words】 Aurora A kinase; endometrial carcinoma; Ishikawa cell 子宫内膜癌是女性生殖系统三大恶性肿瘤之一,约占女性癌症总数7%,占女性生殖道恶性肿瘤20%～30%,近年来子宫内膜癌在世界范围内发病率有上升趋势。在某些欧美国家子宫内膜癌发病率已居女性生殖道恶性肿瘤首位，其发病机制一直倍受关注。Aurora A 是
