ResearchonRadiosensitivityofnasopharyngealcarcinomacellCNE1inVitrotoinhibitorofPI3K／AktInjectionAbstract[Objective]ToevaluatetheradiosensitivitycausedbyLY294002OnnasopharyngealcarcinomacelllineCNE1andinvestigatethepossiblesensitizationmechanismofLY294002.Itwillprovidethebasisofexploringthetargetofnasopharyngealcarcinomaradiotherapysensitization.[MaterialsandMethods]Theexperimentwasdividedintocontrol(A)group，simpledrug(B)goup，simpleradiation(C)groupandradiationplusdrug(D)group．Thecellsurvivalcurvesweredeterminedbycloningassayandcellclonetechniqueforevaluatingthecelllethaleffectsandbymultitargetone-litmodelandL-Qmodel.ThecellcyclesandcellapoptosisrateswereanalyzedbyusingFCM(Flowcytometry)inthedifferentgroupswhichweregivensingledose2Gyradiation．TheBad，Ku-70，Ku80mRNAweredetectedbyRT-PCRmethod，andTheexpressionofpAktandtotalBadweredetectedbywesternblot.StatisticalanalysiswasdonewithSPSSl2．0andSigmaPlot11.0software．ThedataWasexpressedasmeanl+SD，comparisonamonggroupsWasperformedbyOne-WayANOVA，t-testandpearsonlinearcorrelation．SignificanceWasacceptedwhenp0.05．[Results]ExperimentresultofradiosensitvityindicatedthattheabmtyofcelllineCNE1formingcolonyinthetesttinggroupdegradedcontrastingwithAgroup．Withtheincreasingofradiationdose，formingcolonyabilitydecreasedobviously．Withthesameradiationdose，colonyrateintheDgroupwasmuchlessthanthatintheBgroup．TheDoandDqvalueswere0.783Gyand2.54GyintheBgroup，andintheDgroup，theDoandDqwere0.522Gyand2.16Gy.TheSERDowasl.36．a/βvaluewere1.78Gyand9.65Gyrespectively,LY294002canincreasetheradiosensitizationofcelllineCNE1.ThemeasureresultofcellcyclesbyusingFCMdisplayedthatthepercentageinG2/MphaseincreasedafterCNE1cellwasgivenbyLY294002aswellassingledose2Gyradiation．ThecellcyclesinG2/Mphasewereblockedsignificantly(p0.05)intheDgroup.ApoptosisratesintheCgroupWashigherthanthatintheBgroup，whichwas4.63timesand3.54timesthanthatintheAgroup．ApoptosisratesintheDgroupwasthehightestandwas6.33timesthanthatintheAgroup(p0.05)．statisticalanalysisdemonstratedthatthepercen
