食品中不同类型的甜味剂HPLC-ELSD分析方法的研讨

摘 要 摘 要 本学位论文共分为四章。 第一章,概括了甜味剂的现状及发展方向,重点介绍了甜味剂的种类和一些常用甜 味剂的性质与用途,以及甜味剂的主要分析检测方法。 第二章,建立了 HPLC-ELSD 同时检测奶粉中五种水溶性糖的分析方法,样品提取 净化后,采用碳水化合物色谱柱分离(250 mm×4.6 mm ;5 μm ),乙腈/水等度洗脱,蒸 发光散射检测器检测.结果表明:果糖、半乳糖、蔗糖、麦芽糖和乳糖在 1~10 mg/mL 内呈现良好的线性关系,平均回收率为93.0~109%,RSD 为 0.85%~2.7% 。 第三章,为建立高效液相色谱法同时分离测定无糖食品中多种高倍甜味剂的含量, 样品提取净化后,采用 Waters Atlantis® T3 色谱柱分离(250 mm×4.6 mm ;5 μm )、甲 醇/水梯度洗脱;蒸发光散射检测器检测。结果表明:安赛蜜、糖精钠、甜蜜素、三氯 蔗糖、阿斯巴甜进样量 0.5~10 mg/mL 的范围线性良好;RSD4% ,回收率平均在 87.0%~105.0%之间。 第四章,为建立高效液相色谱法同时分离测定无糖食品中多种糖和糖醇的含量,样 品提取净化后,采用高效糖分析色谱柱分离(250 mm×4.6 mm ;5 μm )、乙腈/水梯度洗 脱;蒸发光散射检测器检测。结果表明:木糖、木糖醇、果糖、山梨醇、葡萄糖、蔗糖、 麦芽糖醇、乳糖、乳糖醇进样量 1~10 mg/mL 的范围线性良好;RSD2% ,回收率平 均在 89.2%~110.9%之间。 关键词 高效液相色谱法 甜味剂 食品 I Abstract Abstract This paper consists of four chapters. In the first chapter of this paper,we summarized the market situation of sweeteners, introduced emphatically the nature of some commonly used sweetener, as well as the main analysis sweeteners detection methods. In chapter two ,a quantitative assay for the determination of sugar, such as fructose, galactose, sucrose, maltose and lactose was established by HPLC. Sugar was extracted and the separation was performed with carbohydrate column, acetonitrile/water as mobile phase, Evaporative Light Scattering detector was adopted. The results showed that the linear relation of fructose,galactose,sucrose,maltose and lactose is well in the range of 1~10 mg/ml, the average recoveries in the 93.0-108.9% ,RSD% 2.7. In chapter there,a quantitative assay for the determination of sweetener was established by HPLC. Sweetener was extracted and the separation was performed with Waters Atlantis® T3
