酶法制备杏仁降血压肽的工艺研究摘要近年来,生物活性肽的研究成为热点。本文充分利用工业生产中产生的杏仁粕中的蛋白质资源,将其用于降血压肽的加工,变废为宝,提升了产品的附加值。本文以杏仁蛋白为原料,采用酶解法制备出高活性的降血压肽,主要结果如下:(1)通过单因素实验和Box-Behnken设计响应面优化试验确定最佳酶解条件:pH值为12.48,温度为56℃,底物浓度为3.5%,酶底比为1.3%,在此最优条件下ACE抑制率为82.89%。(2)采用超声波进行辅助酶解,通过单因素实验和正交试验确定最佳超声处理条件超声时间为30min,超声功率为60%,超声温度为40℃。在此最佳实验方案下,测得抑制率为84.57%。(3)通过超滤确定分子量低于3KDa的ACE抑制肽其抑制率达到86.02%;采用阴阳离子混合床脱盐法,确定杏仁ACE抑制肽的脱盐最佳工艺参数为:酶解pH值为4.5、过柱速度为10倍柱/h、阴阳离子比为3:2;经模拟体外胃肠道消化试验后,杏仁ACE抑制肽的抑制率为48.23%,仍具有较高的活性;以《大豆肽粉国家标准GB/T22492-2008》为参考,确定杏仁ACE抑制肽符合大豆肽粉国家一级标准;氨基酸分析结果显示,杏仁ACE抑制肽中含有17种氨基酸,包括8种人体所必需的氨基酸。其中,谷氨酸、天冬氨酸含量最高,分别为12.12%和7.50%;通过SDS-聚丙烯酰胺凝胶电泳法测得杏仁ACE抑制肽的分子量主要集中在2KDa以下。通过上述试验制备出具有降血压肽,具有较高的活性,同时也为杏仁降血压肽工业化生产提供一定的理论依据。关键词:杏仁蛋白;ACE抑制肽;酶法制备;水解度;超滤;脱盐StudyontechnologyofenzymaticpreparationofantihypertensivepeptidesfromAlmondAbstractInrecentyears,bioactivepeptidesbecamethefocusofresearch.Thispapermadefulluseofindustrialproductionofalmondmealproducedtheproteinresources,reuse,enhancetheaddedvalueoftheproducts.WithAlmondproteinasrawmaterialinthispaper,usingenzymehydrolysistopreparedtheantihypertensivepeptidewithhighactivity,themainresultsareasfollows:BysinglefactorexperimentandBox-Behnkendesignresponsesurfaceoptimizationexperimenttodeterminetheoptimalenzymesolutionconditions:pHvaluewas12.48,temperaturewas56℃,substrateconcentrationwas3.5%,enzymeconcentration[E]/[S]%was1.3%,andinthebestconditionACEinhibitionwas82.89%.Usingultrasonicaidedenzymatichydrolysis,throughthesinglefactorexperimentandorthogonaltesttodeterminetheoptimalultrasonictimefor30min,ultrasonicpowerfor60%,ultrasonictemperaturewas40℃,andinthebestconditionACEinhibitionwas84.57%.Throughtheultrafiltrationtodeterminemolecularweightsbelow3KDaofACEinhibitorypeptidesitsinhibitionratereach86.02%;Usingcationexchangeresinsandanionexchangeresinsmixedbedtodesalt,determinedthebestprocessparametersfordesaltingofACEinhibitorypeptides:pHvalueofhydrolyte4.5,hydrolyteflowrate10BV/handbetweenanionexchangeresinsandcationexchangeresins3:2;theinhibitionofACEinhibitorypeptideswas48.23%after
