VII VII undergoing tissue expansion. Histological analysis as Ki-67 immunohistochemistry stain was carried out on the skin samples. Furthermore, microarray analysis was performed to investigate the genome-wide expression profile from about 25,000 human genes. Expression changes of selected genes were verified by RT-PCR on mRNA level. Finally, the potential mechanism of stretch-induced skin regeneration will was discussed. The results were as follows: Histological analysis exhibited normal constitution of expanded skins, and Ki-67 immunohiastochemistry stain manifested enhanced proliferation of epidermal cells in the expanded skins compared with normal skins. Microarray analysis displayed a total of 77 genes that were differentially expressed in expanded skins according to DiffScore standard that is more than 20 or less than -20, with 57 upregulated and 20 downregulated. Another total of 48 genes with more than 2-fold upregulated expression were selected for further discussion, which related to several classes in the context of known biological pathways such as cytokine and chemokine, Cytoskeleton organization, Signal transduction, transcription factors, and so on. In these differentially expressed genes, HOXA5, which VIII VIII belongs to the class of transcriptional factors exhibited the most significant upregulation in all these differentially expressed genes. In addition, HOXB2 was also found overexpressed. The verification by RT-PCR analysis proved consistent results with that of Microarray analysis. The conclusion of our study was explained as follows: Tissue expansion induces the proliferation of epidermal cells, which continued even at the 7th day after the expansion process was stopped. Microarray analysis demonstrated that stretch-induced skin regeneration included a sophisticated network of several integrated cascades, which could be dominated by the most significantly upregulated gene of HOXA5.
