浙江大学硕士学位论文The research about the relationship between myosin light chain kinase expressionand inflammatory bowel disease activityCollege of Medicine，Zhejiang university Internal Medicine(Gastroenterology) Postgraduate Grade 2004：Li jiang bo Supervisor：Gao minAbstractBackground：The intestinal epithelial barrier is frequently disrupted in inflammatory bowel disease(IBD)and this has been proposed to play a role in disease pathogenesis and reactivation．In vitro studies show that cytokine-induced epithelial barrier dys缸ncⅡon Can be mediated by increased myosin light chain kinase (MLCIq expression．Aprimaryfunctionofintestinal epitheliaisthemaintenance ofabarriertopreventunregulatedmixingofluminal and interstitial compartments．While the relevancB ofepifuelial barrier dysfunction todisease pathogenesis remains a subject ofactive investigation，it is well accepted that compromised barrier funetion is a comolon feature ofinfectious，isehemic，and immune-mediated intestinal disease．Objective：To discass myosin light chain expression in inflammatory bowel disease．Methods：Biopsy specimenswereobtainedfrom patients undergoingcolonoscowforIBD，10from ulcerative colitis and 10 from Crohn’s disease．Control samples were obtained from patients undergoing colonoscopy∞part ofa routine program ofpolyp surveillance．The number ofcontrol samples is 10．Then to detect the myosin light chain expression，the above samples are in immunohistochemieal sXaining．Atlastwecalculatethe rateofpositive cellsthroughthemicroscope．Results：The rate ofpositive cells in Crolm’s disease is 50．O％±10．O％．the rate ofpositivecells in ulcerative colitis is 57．6％±25．7％，t11e rate ofpositive cells in conWol samples is 9．2％± 2．7％．the difference between Crohn’s disease and control samples is significunL。(PO．01)so is the difference beween ulcerative colitis and ulcerative colitis．(P0．01)Conclusion：epithelial myosin light chain kinase expression is upregulated in inflammatory bowel disease．Key words：myosin light chain kinase
