维普资讯 · 1488· 理生理杂志 ChineseJournalofPathophysiology 2007,23(8)：1488-1494 [ArticleID] 1000—4718(2007)08—1488—07 Construction ofthevectorforfusion proteingenedriven byIGF -—IIP3 promoteranditsexpressionin hepatocellularcarcinomacells ZHOU Hong—ke，YANG Dong—hua△ TANG Shao—hui，HUANG Wei ， (DepartmentofGastroenterology，TheF ￡AffiliatedHospital，JinanUniversity， Guangzhou510632，China．E—mail：thdyang@163．con) [ABSTRACT] AIM：Toconstructtheshuttleplasmidvectorforthymidinekinase(tk)andEGFPfusionprotein genedrivenbyIGF— IIP3promoter，andinvestigatethespecifickillingeffectoftheHSV—tk／GCV systemonhepatocel— lularcarcinoma(HCC)cellsinvitro．METHODS：Recombinantshuttleplasmidvectorwasconstructedbytechniquesof geneticrecomb inationnadscreening，andidentifiedbyrestrictiondigestionna dsequencinganalysis．Thentherecombinnat shuttleplasmidwas trnasfeetedintoHepG2nadHeLacellsbytechniquesoflipofectaminetrnasfectionna ditsexpression was detectedbyfluorescencemicroscope nadRT—PCR．Cellkillingafterganeielovir(GCV)applicationwasdeterminedby MTr．RESULTS：IdentificationofpDC316一tkEGFP—P3byenzymedigestionandsequencinganalysisshowedthatthe length，insertedlocationnaddirectionofthetargetgeneswhichwereinsertedintotherecombinantwerecorrect．Itwas foundthatenhancedgreenfluorescenceproteincouldonlybeseeninHepG2cells，butnotinHeLacells．TheresultsofRT — PCR showedthatonlytwobna dscouldbe seeninthesamplesofpDC316一tkEGFP—P3trnasfeetedHepG2cells．Th e MTrtestshowedtheselectivecytotoxicityofGCV tothetrnasfeetedHepG2 cells．CONCLUSION ：Th eshuttleplasmid vectorcarryingthetkEGFPfusionproteingenedrivenbyIGF—IIP3promoterhas been
