Thereisevidencethattheplatelet-activatingfactorreceptor(PAFR)isinvolvedintheclearanceofapoptoticcellsbymacrophages, andthatthisisassociatedwithanti-in?ammatoryphenotype.Ourgrouphaspreviouslyshownthatcoinjectionofalargenumber ofapoptoticcellscanpromotetumorgrowthfromasubtumorigenicdoseofmelanomacells.Here,westudiedtheinvolvement ofthePAFRinthetumorgrowthpromotinge?ectofapoptoticcells.Asub-tumorigenicdoseofmelanomacells(Tm1)was coinjectedwithapoptoticTm1cells,subcutaneouslyinthe?ankofC57Bl/6mice,andthevolumewasmonitoredfor30days. AnimalsreceivedthePAFRantagonists,WEB2170orPCA4248(5mg/kgbodyweight)orvehicle,byperitumoraldailyinjection for5days.ResultsshowedthatPAFRantagonistssigni?cantlyinhibitedthetumorgrowthinducedbythecoinjectionofasub- tumorigenicdoseofmelanomacellstogetherwithapoptoticcells.Thiswasaccompaniedbyinhibitionofearlyneutrophiland macrophagein?lt
