Asimple,sensitive,andrapidHPLC-MS/MSmethodwasdevelopedandvalidatedforquantitativeestimationofmemantinein humanplasma.ChromatographywasperformedonZorbaxSB-C18(4.6×75mm,3.5μm)column.Memantine(ME)andinternal standardMemantine-d6(MED6)wereextractedbyusingliquid-liquidextractionandanalyzedbyLC-ESI-MS/MSusingmultiple- reactionmonitoring(MRM)mode.Theassayexhibitedalineardynamicrangeof50.00–50000.00pg/mlforMEinhumanplasma. Thismethoddemonstratedanintra-andinterdayprecisionwithintherangeof2.1–3.7and1.4–7.8%,respectively.Furtherintra- andinterdayaccuracywaswithintherangeof95.6–99.8and95.7–99.1%correspondingly.ThemeanrecoveryofMEandMED6 was86.07±6.87and80.31±5.70%,respectively.ThedescribedmethodwassuccessfullyemployedinbioequivalencestudyofME inIndianmalehealthyhumanvolunteersunderfastingconditions. 1.Introduction
