SleepPhenotypinginaMouseModelofExtremeTrait Anxiety VladimiraJakubcakova,CorneliaFlachskamm,RainerLandgraf,MayumiKimura* MaxPlanckInstituteofPsychiatry,Munich,Germany Abstract Background:Thereisaccumulatingevidencethatanxietyimpairssleep.However,duetohighsleepvariabilityinanxiety disorders,ithasbeendifficulttostateparticularchangesinsleepparameterscausedbyanxiety.Sleepprofilinginananimal modelwithextremelyhighvs.lowlevelsoftraitanxietymightservetofurtherdefinesleeppatternsassociatedwiththis psychopathology. Methodology/PrincipalFindings:Sleep-wakebehaviorinmouselineswithhigh(HAB),low(LAB)andnormal(NAB)anxiety- relatedbehaviorswasmonitoredfor24hduringbaselineandrecoveryafter6hsleepdeprivation(SD).Theamountsof eachvigilancestate,sleeparchitecture,andEEGspectralvariationswerecomparedbetweenthemouselines.Incomparison to NAB mice, HAB mice slept more and exhibited consistently increased delta power during non-rapid eye movement (NREM)sleep.Theirsleeppatternswerecharacterizedbyheavyfragmentation,reducedmaintenanceofwakefulness,and frequentintrusionsofrapideyemovement(REM)sleep.Incontrast, LABmiceshowedarobustsleep-wakerhythmwith remarkably prolonged sleep latency andalong, persistent periodof wakefulness. Inaddition, the accumulationof delta powerafterSDwasimpairedintheLABline,ascomparedtoHABmice. Conclusions/Significance:Sleep-wakepatternsweresignificantlydifferentbetweenHABandLABmice,indicatingthatthe geneticpredispositiontoextremesintraitanxietyleavesabiologicalscaronsleepquality.Theenhancedsleepdemand observed in HAB mice, with a strong drive toward REM sleep, may resemble a unique phenotype reflecting not only elevatedanxietybutalsoadepression-likeattribute.
