肝癌大鼠体内组织因子表达水平的研究学位申请人：陈浩导师：金毕 教授摘 要目的：通过直接注射法建立大鼠肝癌模型，同时设立对照组，观察两组大鼠血浆内组 织因子表达水平的变化并加以比较，从而了解在肝癌大鼠体内组织因子的表达水平。 方法：选用 Walker-256 榴株作为肝癌细胞株，利用 Wistar 大鼠进行腹腔内传代，一周 后得到癌性腹水离心加以浓缩。其中一组 SD 大鼠手术暴露肝脏后采用穿刺注射方法 种植肿瘤以建立肝癌模型，另一组 SD 大鼠作为空白手术对照。分别于手术前一周、 手术后一周、手术后两周采集大鼠血清样本，并用酶联免疫吸附实验测定其血浆中组 织因子的含量。结果：与术前相比，两组大鼠在术后 7d 血浆中组织因子的表达水平较前均有轻微升 高，差异并无统计学意义（P0.05）。术后 14d，肝癌组大鼠血浆中组织因子的表达 水平显著升高，明显高于同时期对照组以及术前和术后 7d 水平（P0.05），差异具 有统计学意义；对照组大鼠血浆中组织因子的表达水平与术前及术后 7d 的表达水平 并无明显差异(P0.05)。结论：肝癌大鼠体内组织因子的表达水平有明显的升高。 关键词：组织因子；肝癌；大鼠模型A research of tissue factor expression level of liver cancer in ratsCandidate:Hao Chen Supervisor:Bi JinABSTRACTObjective: We established a rat model group of liver cancer by direct injection and set up a control group, to observe and compare the expression level of tissue factor of the two groups, in order to know the expression level of tissue factor in rats with liver cancer. Methods: We chose Walker-256 cell line as the cell line of liver cancer, then we used Wistar rats for intraperitoneal passage of the tumor cell line, the ascites was extracted from abdomen after one week and then was concentrated by centrifugation. One group of rats was operated through the exposure of liver and the injection of the concentrated ascites to be set as a cancer model group, while the other group was set as a blank contrast. We collected blood samples from both group of rats one week before, one week after and two weeks after the operation, the sample are tested by enzyme-linked immunosorbent assay to determine the expression level of tissue factor.Results: Comparing to the data before the surgery，levels of tissue factor expression in bothgroups rose slightly after one week of the operation; up to two weeks the average expression level of the cancer model group is significantly higher than the level of before and the control group, while the level of the control group does not present such a change.
