中 文 摘 要莲房是睡莲科植物莲 Nelumbo nucifera Gaertn.的干燥花托，为《中国药典》 2005 版一部所收载。文献报道莲房中富含原花青素，原花青素具有清除自由基、 抗氧化等作用，作为原料或添加剂已广泛应用于医药保健品、化妆品、营养补充 剂等产品。为进一步扩大原花青素原料来源，开发利用莲房资源，本课题针对莲 房药材质量控制方法及原花青素提取物生产工艺进行了系统研究。主要研究内容 及结果如下：一、药材质量控制方法研究莲房药材质量控制方法《中国药典》2005 版项下仅规定了其来源、性状、 显微和理化鉴别，及水分和总灰分检查。本研究首次对莲房化学成分进行了系统 的分离鉴定，并建立了莲房药材定性定量方法，可为药材标准完善提供依据。1.莲房化学成分研究 以采自湖南省湘潭市的莲房为原料，应用系统溶剂分 离法分离出化合物 A。经波谱鉴定并与已知品对照，鉴定为金丝桃苷（3-O-?-D- galactopyranosyloxy-2-(3,4-dihydroxy phenyl)-5,7-dihydroxy-4H-1-benzopyran-4-one）。2．莲房药材定性方法研究 首次以金丝桃苷为对照，对 4 批莲房药材样品进 行了 TLC 定性鉴别，结果斑点清晰，分离度好，可作为莲房药材的定性鉴别方 法。3．莲房药材定量方法研究 首次以金丝桃苷为对照，采用紫外-可见分光光 度法测定药材中总黄酮含量，RP-HPLC 法测定药材中金丝桃苷的含量。两种方法 均简便，重复性好，可作为莲房药材的质量控制方法。 二、莲房原花青素的提取纯化工艺研究有关莲房中原花青素的提取纯化工艺未见报道，本试验采用乙醇回流提取、 大孔吸附树脂纯化，首次对工艺参数进行了系统研究。1．莲房原花青素提取工艺优化 以提取液中原花青素含量为指标，对提取溶 媒、乙醇浓度和提取温度进行了单因素考察。在此基础上以 L9 (34 )正交表设计试验，以浸出物得率、原花青素含量为评价指标，考察了乙醇用量、提取时间、提取次数等影响因素，结果以 10 倍量 60%乙醇 80℃回流提取 2 次，每次 1 小时 为优。2．大孔吸附树脂纯化莲房原花青素 以原花青素的静态吸附率与解吸率为指 标，考察了 4 种大孔吸附树脂对原花青素的吸附性能，以优选的树脂型号考察了 影响树脂动态吸附与解吸相关因素。研究表明：采用 AB-8 大孔吸附树脂，吸附 阶段药液浓度 0.2g(生药)/ml 为佳；解吸阶段以蒸馏水 4BV、30%乙醇 4BV 依次 洗脱，收集 30%乙醇洗脱部位，回收溶剂，即得莲房原花青素。拟定的工艺简单、 易控，莲房原花青素提取物可达到较好的纯化效果，原花青素的精制度达到 170.18％，纯度达 59.85％，固形物收率达 46.47％。 关键词 莲房 金丝桃苷 原花青素 制备工艺 药材质量控制 AbstractLianfang，which is recorded in the 2005 edition of Chinese Pharmacopoeia, is the dry receptacle of Nelumbo nucifera Gaertn..
