第五部分M5以及M6体内应用的安全性研究【目的】探讨M5以及M6在体内应用的安全性问题【方法】分别将PBS，Ad5/dE1A，M5，M6以及Adv-TK注射入小鼠体内，取眼动脉血进行肝肾功能检测，HE染色检测各脏器病理反应，real-timePCR法检测各组织腺病毒的表达情况，流式细胞术检测外周血以及脾脏中CD4+以及CD8+淋巴细胞的变化情况,TCID50法检测血清中抗腺病毒中和抗体。【结果】同PBS组相比，M5，M6以及Adv-TK仅引起一过性的肌酐的升高；小鼠的肝，脾，肾等也未见明显的病理改变；M5，M6以及Adv-TK被注射入体内后，在肝，脾，肾中均有表达，其中在肝中表达的时间较长，且三种病毒之间的表达无明显差异；外周血中的CD4+以及CD8+淋巴细胞在病毒注射后无明显变化，而脾中的CD4+以及CD8+淋巴细胞均出现一过性的减少，且M5，M6以及Adv-TK引起的变化趋势以及程度相似；M5，M6以及Adv-TK在小鼠中引起的中和抗体的数量以及变化趋势无明显差异。【结论】M5以及M6在体内应用具有较好的安全性【关键词】安全性中和抗体淋巴细胞腺病毒EnglishAbstractPartⅠTheconstructionandidentificationofnewoncolyticadenovirusM5andM6【Objective】ToconstructionandidentificationoftwooncolyticadenoviruswhichexpressHSV-TKgene【Methods】HSV-TKgenewasacquiredbyPCRandlinkedtoPcDNA3.1+/E3⊿6.7k/gp19kandPcDNA3.1+/E3⊿ADP,M5andM6wereconstructedbyhomologousrecombination.HirtDNAofM5andM6wasacquiredfromtheinfected293cells,thenPCRandenzymecutwereusedtoidentificationM5andM6.【Results】ThesequenceofPCRproductwasidentifiedtoberightbysequencingcompany,PCRwasdoneusingthetemplateofvirusDNA,andtheproductwasseparatedona1.0%agarosegel,abondwasseeninaround1.2kb,virusDNAwasenzymecutbyBanⅢandtheproductwasalsoseparatedona0.8%agarosegel,andthepurpusebandswereseeninaround1.2kbandabove30kb.【Conclusion】oncolyticadenoviruswhichexpressHSV-TKgenewereconstructedsuccessfully.【Keywords】oncolyticadenovirushomologousrecombinationenzymecutPartⅡTheinvitroeffectcomparationofoncolyticadenovirusM5,M6andreplication-deficientadenovirusAdv-TK【Objective】TocomparetheinvitroeffectofoncolyticadenovirusM5,M6andreplication-deficientadenovirusAdv-TK【Methods】AftertumorcellsandnormalcellswereinfectedwithM5,M6andAdv-TK,theexpressionofTKproteinwasdetectedbyWesternblot,theCPEeffectwasdetectedbycrystallizationvioletstain,thereplicationablityintumorcellsandnormalcellswasdetectedbyrealtimePCR,thelethaleffecttonormalcellsandtumorcellswasdetectedbyMTTandflowcytometry.【Results】Intumorcells,theTKproteinexpressionofM5andM6washigherthanAdv-TK,andtheexpressionofM6wasthehighest,however,innormalcells,theTKproteinexpr
