狼毒粉狼毒大枣汤单次给药毒性研究 　　[摘要] 目的 研究狼毒粉（EFSP）、狼毒大枣汤（DEFSJ）的单次给药毒性反应，为安全用药提供参考。 方法 分别制备EFSP、DEFSJ，采用改良寇式法测定其半数致死量（LD50）及其95%可信度；并通过检测小鼠血液生化学和组织病理学检查评价其对小鼠肝、肾功能的影响。 结果 EFSP的LD50为2.12 g/kg，95%可信限为1.61～2.78 g/kg，DEFSJ的LD50为12.60 g/kg，95%可信限为9.65～16.48 g/kg。EFSP、DEFSJ给药组与枣汤组比较，随着给药剂量的增加，体重呈显著降低趋势，但脏器系数变化不明显；小鼠血液生化学结果显示，血清中谷丙转氨酶、谷草转氨酶水平均升高（P 0.05），肌酐、血尿素氮水平有升高的趋势；病理学检查结果显示，两药对小鼠的肝、肾功能有一定的损伤。 结论 EFSP、DEFSJ均有一定的肝、肾毒性。 　　[关键词] 狼毒粉；狼毒大枣汤；改良寇式法；单次给药毒性 　　[中图分类号] R285.5 [文献标识码] A [文章编号] 1673-7210（2018）01（b）-0004-06 　　[Abstract] Objective To study single drug toxicity of Euphorbia fischeriana Steud Powder （EFSP） and Decoction of Compatibility of Euphorbia fischeriana Steud and Jujubae （DEFSJ） and provide a reference for safe medication. Methods Modified Karber method was adopted to detect lethal dose 50 （LD50） and the 95% confidence interval of EFSP and DEFSJ. Blood biochemistry and histopathology were detected to evaluate the toxicity of EFSP and DEFSJ on liver and kidney of mice. Results The LD50 of EFSP was 2.12 g/kg， and the average 95% confidence interval was 1.61-2.78 g/kg. The LD50 of DEFSJ was 12.60 g/kg， and the average 95% confidence interval was 9.65-16.48 g/kg. Compared with the Jujubae Decoction group， as the dosage was increased， the mice weight was significantly decreased， however， organ coefficient was not obviously changed in EFSP and DEFSJ groups. The blood biochemical results showed that the levels of alanine transaminase and aspartate transaminase were elevated （P 0.05）， and the levels of creatinine and blood urea nitrogen had an increased tendency. Pathological changes of kidney and liver were found by histopathological examination.
