肿瘤放射抗拒性的研究进展 摘要 ：肿瘤是我国常见恶性疾病之一，放射治疗是其重要的治疗手段之一，放疗后复发与 转移导致了放射治疗的失败， 而肿瘤干细胞的放射抗性在其中扮演着重要角色。 肿瘤干细胞 假说是近年来提出的一种新理论， 具有自我更新、 多向分化等多种潜能， 是肿瘤预后不良的 一个重要因素， 此概念的提出为研究肿瘤的发生、 发展、 治疗提供了新视野。肿瘤干细胞放 射抗性的相关机制错综复杂，其中涉及凋亡基因的调控、 DNA损伤修复能力、细胞周期状态 以及多种分子信号通路的改变。 文章对肿瘤干细胞的起源、 肿瘤干细胞放射抗拒生物特性和 相关机制及其临床意义 , 研究前景进行了阐述。 关键词：肿瘤干细胞；放射抗拒 Abstract: Tumor is one of the most common malignant diseases, radiation therapy is one of the important measures of treatment, recurrence and metastasis after radiotherapy cause the failure of the radiotherapy, radiation resistance and cancer stem cells play an important role in it. Cancer stem cell hypothesis is a new theory put forward in recent years. Cancer stem cell has the potential of self-renewal and multi-directional differentiation, which is an important factor of tumor prognosis.The concept is provides a new perspective for the research, development and treatment of tumor. Radiation resistive mechanism of tumor stem cells is complex, involving apoptosis gene regulation, DNA damage repair ability, the change of the state of the cell cycle as well as a variety of molecular signaling pathways. This article will show that recent origin, radiation resist biological characteristics, mechanism, clinical significance and research prospects of cancer stem cells. Key words: cancer stem cells; radiation resistance; molecular signaling pathways 1 肿瘤干细胞的起源 目前存在两种肿瘤形成学说，即随机学说与阶层学说 [1] 。随机学说认为所 有肿瘤细胞是功能同质的， 每个肿瘤细胞都有形成肿瘤的能力， 但其进入细胞周 期增殖分裂是由一些低概率的随机事件控制。 因此，要阐明肿瘤的生物学特性必 须研究全部肿瘤细胞。而阶层学说认为，肿瘤细胞在功能上存在很大的异质性， 只有极小部分肿瘤起源细胞才有成瘤能力， 且成瘤率很高。 这种肿瘤起源细胞即 为肿瘤干细胞， 与其他肿瘤细胞不同， 它具有自我更新和分化能力，
