Systemiclupuserythematosus(SLE)isaprototypicin?ammatoryautoimmunedisordercharacterizedbymultisysteminvolvement and?uctuatingdiseaseactivity.Symptomsrangefromrathermildmanifestationssuchasrashorarthritistolife-threateningend- organmanifestations.DespitenewandimprovedtherapyhavingpositivelyimpactedtheprognosisofSLE,asubgroupofpatients donotrespondtoconventionaltherapy.Moreover,theriskoffataloutcomesandthedamagingsidee?ectsofimmunosuppressive therapiesinSLEcallforanimprovementinthecurrenttherapeuticmanagement.NewtherapeuticapproachesarefocusedonB- celltargets,T-celldownregulationandcostimulatoryblockade,cytokineinhibition,andthemodulationofcomplement.Several biologicalagentshavebeendeveloped,butthisencouragingnewsisassociatedwithseveraldisappointmentsintrialsandprovide atimelymomenttore?ectonbiologictherapyinSLE. 1.Introduction molecules[4].Besideautoantibodyproduction,B-cellsare thekeyfortheactivationoftheimmunesystem,particularly throughcytokinesandasantigen-presentingcells.Animpor- tantpartofB-cellsisactivatedinaT-cell-dependantmanner.
