(Equal participation of KZ and EM) InstitutNationaldelaSant′eetdelaRechercheM′edicale(INSERM)U331,Facult′edeM′edecineRTHLa¨ennec,Lyon,France CD36,an88kd-adhesionmolecule,playsamajorroleasascavengingreceptorimplicatedincellularlipidmetabolism.Secretory mammaryepithelium,microvasculatureendothelium,adipocytes,smoothmusclecells,andplateletsexpressCD36.Inaddition, CD36expressionissigni?cantlyenhancedinmacrophagesdi?erentiatingintofoamcells.Thee?ectofpathologicallevelsofcholes- terol,asobservedinapoE?/?,onvascularCD36expressionis,atthisstage,notknown.Inthisstudy,aquantitativeanalysisofCD36 transcriptionandproteinexpressionlevels,presentintissuesofmaleC57BL/6andapolipoprotein-E(apoE)de?cientmicewas carriedoutbyNorthernandWesternblots.Four-week-oldanimalswerefedachowdietoverdi?erentperiodsoftime(0,6,16, or20weeks).ImmunohistochemistrywasusedtolocalizeCD36proteinexpressionintheheartandtestis.Resultsindicatethat CD36transcriptionisincreasedinheartsofapoEde?cientanimals(100%higherat6weeks,and30%higherat16and20weeks) incomparisontowildtype.Thiswascon?rmedattheproteinlevel,whichshowedanincreaseofatleast100%at6weeks,and between40%to50%increaseat16and20weeksofapoE?/?micecomparedtocontrols.Inaddition,CD36transcriptionlevelswere signi?cantlyincreasedintestisofapoEanimals(atleast100%at6,16,and20weeks)comparedtoC57BL/6wildtype.Suchan increasewasalsocon?rmedattheproteinlevel(65%increaseat16weeksinapoEmicecomparedtocontrol).Finally,localization ofCD36proteinexpressionbyimmunohistochemistryshowedthatitwasexpressedinthecapillariesofheartandtestisendothelial cellsandalsoattheheadofspermatozoidduringspermatogenesis.Theseresultsindicatethathighcirculatingcholesterollevels,in apoEde?cientmice,signi?cantlyenhancetheexpressionofCD3
