Somaticcellsshowaspontaneousdeclineingrowthrateincontinuousculture.Thisisnotrelatedtoelapsedtimebuttoan increasingnumberofpopulationdoublings,eventuallyterminatinginaquiescentbutviablestatetermedreplicativesenescence. Thesecellsarecommonlymultinucleatedanddonotrespondtomitogensorapoptoticstimuli.Cellsdisplayingcharacteristics ofsenescentcellscanalsobeobservedinresponsetootherstimuli,suchasoncogenicstress,DNAdamage,orcytotoxicdrugs andhavebeenreportedtobefoundinvivo.Mosttumorsshowunlimitedreplicativepotential,leadingtothehypothesisthat cellularsenescenceisanaturalantitumorprogram.Recent?ndingssuggestthatcellularsenescenceisanaturalmechanismto preventundesiredoncogenicstressinsomaticcellsthathasbeenlostinmalignanttumors.Giventhattheultimategoalofcancer researchisto?ndthede?nitivecureforasmanytumortypes aspossible,explorationofcellularsenescencetodrivetowards antitumortherapiesmaydecisivelyin?uencetheoutcomeofnewdrugs.Inthepresentpaper,wewillreviewthepotentialof cellularsenescencetobeusedastargetforanticancertherapy. 1.TheBiologyofSenescence cellular lifespan with metabolic
