Pluripotentstemcellsareregardedasapromisingcellsourcetoobtainhumandopamineneuronsinsu?cientamountsand purityforcellreplacementtherapy.Importantly,thesuccessofclinicalapplicationsdependsonourabilitytosteerpluripotent stemcellstowardstherightneuronalidentity.InParkinsondisease,thelossofdopamineneuronsismorepronouncedinthe ventrolateralpopulationthatprojectstothesensorimotorstriatum.Becausesynapsesarehighlyspeci?c,onlyneuronswiththis preciseidentitywillcontribute,upontransplantation,tothesynapticreconstructionofthedorsalstriatum.Thus,understanding thedevelopmentalcellprogramofthemesostriataldopamineneuronsiscriticalfortheidenti?cationoftheextrinsicsignalsand cell-intrinsicfactorsthatinstructand,ultimately,determinecellidentity.Here,wereviewhowextrinsicsignalsandtranscription factorsacttogetherduringdevelopmenttoshapemidbraincellfates.Further,wediscusshowthesesamefactorscanbeappliedin vitrotoinduce,select,andreprogramcellstothemesostriataldopaminefate. 1.TheCentralRoleofVentralMidbrain DopamineNeuronsinParkinsonDisease lack Lewy bodies [5–7] whilst, on the other hand, Lewy bodiescanbefo
