敦煌平胃丸对胃癌前病变大鼠Notch信号通路中Notch2和Jagged1表达影响 　　【摘要】目的：观察敦煌平胃丸对胃癌前病变（PLGC）模型大鼠胃黏膜组织Notch信号通路中Notch2和Jagged1表达的影响。方法：采用复合法建立PLGC大鼠模型，将32只大鼠随机分为4组，每组8只。敦煌平胃丸组用敦煌平胃丸浸膏按每日生药96 g / kg掺入饲料中喂养给药，共计12周。观察大鼠胃黏膜病理组织学变化；采用Western Blot检测Notch2和Jagged1蛋白表达；采用RT-PCR检测Notch2和Jagged1mRNA的表达。结果：PLGC大鼠胃黏膜组织Notch2 与Jagged1mRNA及蛋白表达较正常大鼠胃黏膜组织均明显上调（P005）。敦煌平胃丸能明显改善PLGC大鼠胃黏膜腺体萎缩、肠化增生，下调PLGC大鼠胃黏膜组织中Notch2 与Jagged1表达水平（P005）。结论：Notch2、Jagged1激活了Notch信号通路，在PLGC发生发展中有重要意义。敦煌平胃丸治疗PLGC可能与下调Notch信号通路中Notch2 与Jagged1表达有关。 　　【关键词】敦煌平胃丸；胃癌前病变；Notch2、Jagged1 　　【中图分类号】R2855【文献标志码】 A【文章编号】1007-8517（2015）01-0018-04 　　Abstract：Objective To observe the Dunhuang pingwei pill on the expression of Notch2 and Jagged1 in the Notch signal pathway in rats with precancerous lesions of gastric cancer.Methods To establish the rat model of PLGC by using composite method， 32 rats were randomly divided into 4 groups， 8 rats in each group， Dunhuang pingwei pill group were fed with Dunhuang pingwei concrete according to daily crude drug 96 g/kg soak in feedstuff ， a total of 12 weeks.Observe changes in gastric mucosa pathological histology in rats.Using Western Blot to detect protein expression of Notch2 and Jagged1， the transcriptions of Notch2 and Jagged1mRNA were demonstrated by RT-PCR technique.Result Compared to the normal rats， Notch2 and Jagged1mRNA、protein expression were significantly up-regulated in gastric mucosa histology in rats with PLGC .（P 005）.Dunhuang pingwei pill can obviously improve the gastric mucosa gland atrophy， intestinal metaphases and hyperplasia ， down-regulate the expression level of Jagged1andNotch2 in gastric mucosa tissue in rats with PLGC .（P 005）.Conclusion Notch2， Jagged1 activates Notch signaling pathways in PLGC， it is important to the development of PLGC.Dunhuang pingwei pill in the treatment of PLGC maybe associated with down-regulating expression of Notch2 and Jagged1 in Notch signal pathway. 　　Keywords：Dunhuang pingwei pill， PLGC，Notch2，Jagged1 　　胃癌前病变（ gastric precancerous
