Abstract Objective: To study the effect of Puerarin on proliferation and apoptosis of human gastric cancer MGC-803 and AGS cells ． New treatment methods for the clinical treatment of gastric cancer. Methods: Human gastric cancer cells were treated with Puerarin at different concentrations（1.5、 3、6、12、24 mmol/l）．MTT assay was used to test cell proliferation and FCM was used to detect cell apoptosis．Application SPSS13.0 statistical software for processing the experimental data are expressed as mean ± standard deviation ( x  ± s ),group pairwise comparisons using one-way ANOVA, betwent the groups were compared using independent-samples t test analysis. P 0.05 considered statistically significant difference. Results: MTT assay groups of cell growth inhibition, the results show that the treatment of gastric cancer cells were treated with different doses of puerarin, subject to varying degrees of cell growth inhibition,and the degree of inhibition is enhanced with the increase of drug concentration. The 48h IC50 values of two gastric cancer cells obtained by the SPSS software were 22.67mmol/l and 16.17mmol/l. FCM analysis to detect the rate of apoptosis of gastric cancer cell MGC-803 control group was 5.49 ± 0.32%, using different concentrations （12、24mmol/l）puerarin for 24h apoptosis rate was 9.53±0.62%,13.81 ± 0.42%,gastric cancer cells AGS apoptosis rate in the control group was 6.23 ± 0.21%, using different concentrations （12、24 mmol/l）puerarin for 24h apoptosis rate was 16.38±1.0%,25.99 ± 0.31%. Dosing group and the control group, the apoptosis rate differences was statistically significant (P 0.05).
