Ab Abstract 万方数据 万方数据 ABSTRACT Objective ： To establish the mouse model of paraquat -induced lung injury , investigate the protective effect of mast cell stabilizer on lung injury induced by paraquat in mice. Methods：Thirty-six female Kunming mice were randomly divided into control group,paraquat group,captopril group and sodium cromolyn group. There were 18 mice in paraquat group,and 6 mice in the rest three groups.Each mouse in control group was injected intraperitoneally 1ml saline,after 0.5h and then every 12h each mouse was injected intraperitoneally 1ml saline till being sacrificed at 72h.Each mouse in PQ group was injected intraperitoneally PQ(by 30mg/Kg body weight standard),after 0.5h and then every 12h each mouse was injected intraperitoneally 1ml saline till being sacrificed ， 6 mice were sacrificed at 8h 、 24h and 72h.Each mouse in captopril group was injected intraperitoneally PQ(by 30mg/Kg body weight standard),after 0.5h and then every 12h each mouse was injected intraperitoneally captopril(by 10mg/Kg body weight standard) till being sacrificed at 72h.Each mouse in cromolyn sodium group was injected intraperitoneally PQ(by 30mg/Kg body weight standard),after 0.5h and then every 12h each mouse was injected intraperitoneally cromolyn sodium(by 25mg/Kg body weight standard) till being sacrificed at 72h.After administration,mice were observed of mental state, hair changes ,behavioral changes and so on.We observed the lungs and put them into 10% neutral . The lungs in PQ group at 8h and 24h were only for HE staining；and the lungs in each group at 72h were for HE staining,Masson staining, COX-2 、 TGF-β1 and CD68 immunohistochemistry. Results ： (1) Mice in PQ group appeared poisoning symptoms after PQ poisoning,and the symptoms gradually increased with time.The poisoning symptoms of mice in captopril group and cromolyn sodium group were reduced compared with PQ group,but there was no significant difference between captopril group and cromolyn sodium group.
